Thidiazuron induced somatic embryogenesis in Coffea arabica L. and Coffea canephora P ex Fr

چکیده
The rapid direct and repetitive somatic embryogenesis in Coffea arabica and C. canephora genotypes was tested on Murashige and Skoog medium (MS) containing thidiazuron (TDZ) (1-phenyl-3-(1,2,3,-thiadiazol-5-yl)urea) in concentrations of 2.27–11.35s M. Segments taken from cotyledon leaf, first leaf and stalk of regenerated plantlets produced clusters of somatic embryos directly from cut portions of explants on TDZ (9.08 M) containing medium within a period of two months. Subculturing of these embryo clusters produced more secondary embryos on reduced TDZ (0.045–0.91 M) containing medium and these subsequently developed into plantlets (80–85%) on development medium followed by rooting on MS basal medium. This direct somatic embryogenesis from leaf and hypocotyl explants in Coffea sp. is a strong evidence of cell totipotency. The rapid somatic embryo induction protocol would be useful for the mass propagation, direct regeneration and genetic transformation of selected elite lines.

Analysis of Phosphorus Use Efficiency Traits in Coffea Genotypes Reveals Coffea arabica and Coffea canephora Have Contrasting Phosphorus Uptake and Utilization Efficiencies
BACKGROUND AND AIMS Phosphate (Pi) is one of the most limiting nutrients for agricultural production in Brazilian soils due to low soil Pi concentrations and rapid fixation of fertilizer Pi by adsorption to oxidic minerals and/or precipitation by iron and aluminum ions. The objectives of this study were to quantify phosphorus (P) uptake and use efficiency in cultivars of the species Coffea arab...

Green coffee beans of Coffea arabica and Coffea canephora accessions from different geographical origin (Brazil, Colombia, Ethiopia, Honduras, Kenya, Mexico, Peru, Uganda and Vietnam) were extracted and the extracts analyzed by HPLC-ESI-MS/MS for the identification and quantification of chlorogenic acids and caffeine content. Principal component and cluster analyses were used to identify chemic...
متن کاملBioreactors in coffee micropropagation
In coffee, bioreactors are the most promising way for scaling-up micropropagation processes, particularly somatic embryogenesis. The availability of an efficient somatic embryogenesis process would allow the rapid mass production of heterozygous materials such as selected Coffea canephora clones and F1 Arabica hybrid varieties. For the last fifteen years, bioreactors (mechanically or pneumatica...
متن کاملCoffeebEST: an integrated resource for Coffea spp expressed sequence tags.
Coffee is one of the most important commodities in the world, and its production relies mainly on two species, Coffea arabica and Coffea canephora. Although there are diverse transcriptome datasets available for coffee trees, few research groups have exploited the potential knowledge contained in these data, especially with respect to fruit and seed development. Here, we present a comparative a...
متن کاملSomatic Embryogenesis: Identified Factors that Lead to Embryogenic Repression. A Case of Species of the Same Genus
Somatic embryogenesis is a powerful biotechnological tool for the mass production of economically important cultivars. Due to the cellular totipotency of plants, somatic cells under appropriate conditions are able to develop a complete functional embryo. During the induction of somatic embryogenesis, there are different factors involved in the success or failure of the somatic embryogenesis res...
